The vascular endothelial growth factor gene (VEGF) promotes angiogenesis and is expressed and detected in cancer lymphoma cells. We report transcription levels of VEGF in spleen of N-methyl-N-nitrosourea (MNU)-induced splenic lymphoma in Sprague Dawley rats through oral and intraperitoneal (IP) administration. Groups of 6 week old female rat not-treated controls (6 rats), treated orally with 60mg/kg MNU (9 rats) or IP with 60mg/kg MNU (9 rats ), were used. MNU was given 4 times in 2 week periods and rats were kept 24 weeks before spleen samples were collected for total RNA extraction. Transcription levels of VEGF transcript were determined using quantitative real-time reverse transcriptase PCR (qRT-PCR). RNAs were converted into cDNA and amplified by real time PCR machine. Results of amplification of VEGF transcripts were normalized with housekeeping genes glyceraldehydes-3-phosphate dehydrogenase (GAPDH) and beta-actin. The relative transcription of VEGF transcript in the spleen of MNU-IP group was 360.38 ± 23.25SD fold or 316 times higher (P < 0.05) than controls (1.14±0.08SD fold), whereas transcription in the MNU-oral group was 44.97±1.07 fold or 39 times higher than controls. The findings suggest that VEGF transcript was highly transcribed in rat spleen exposed to MNU via IP route.
