2013. Valproic acid (VPA) is
an anti-epileptic used to control epilepsy, migraine and bipolar psychiatric
disorders. The effect of daily doses of 100, 200, 400 and 500mg/kg VPA given
intraperitoneally (IP) to 4 groups of 10 mice each for 90 days respectively, on
maturation of oocytes, fertilization and embryonic development in vitro is reported. Controls were
another group 10 mice injected IP with saline. Oocyte maturity and
fertilization were assessed 24 h after the last dose and the number of
apoptotic cells in blastocyst was evaluated by TUNEL assay. Miosis II (MII),
oocyte maturation, fertilization and blastocyst rate were significantly lower
in mice given VPA compared with controls. The effects were dose dependent.
Chronic administration of VPA increased apoptotic cells in blastocysts and
induced early embryonic death. The findings suggest that chronic IP
administration of VPA in IPC mice could adversely affect pre-implantation of
embryo development and reduce fertility.
