 Effect of Trigonella seed extract on ischemic injury in male Wistar rats.

We report effect of Trigonella seed extract in groups of 6 male 280-300g Wistar male rats each with (1) or without (2) renal ischemia-reperfusion or gavaged 0.3g extract (3) daily for 14 days. Controls (1) underwent ischemia but were given saline only. Saline (controls) and Trigonella extract were gavaged daily 14 days before clamping aortic renal artery at day 14 for 45min, then unclamped for reperfusion for 1h. At day 15, left kidneys were removed and tissues subjected to microscopy. In rats given saline but clamped, we observed severe vacuolar degeneration, necrosis of proximal and distal tubules, cellular swelling, foamy cytoplasm, detachment of epithelial cells, nuclear pyknosis, glomerular atrophy, dilation, congestion and hemorrhage. Lesions were scored 0-5 and compared (P < 0.05) with unclamped saline controls, in clamped rats, tubular degeneration scored 3-4, tubular necrosis 2-3 and histopathology 3.5-5. However, in ischemic rats gavaged Trigonella we found only few proximal tubules exhibiting mild epithelial changes and tubular degeneration scored 1-2, tubular necrosis 0-2 and histopathology 0.5-2.5 (P < 0.05).
