2017. The anti-allergic drug Tranilast may induce cell line
proliferation and prevent apoptosis. MCF-7 cells were grown in Dulbecco's modified eagle's
medium, fetal bovine serum, non-essential amino acid,
L-glutamine, penicillin and streptomycin in T-25 cm2 tissue culture
flasks. Cells were plated in 24 well culture plates with 104
cells/mL densities for 12h, further cultured in Tranilast
media for 24h. Percentage viability was measured at 490-492nm by ELISA. Morphine was used as an apoptosis control. One day after
plating, cells were washed with PBS. Effect of Tranilast
on viability and cell death was determined by microscopy, trypan blue, Hoescht/PI staining and lactate
dehydrogenase test. Data analyzed using one-way ANOVA followed by Tukey’s post
hoc tests suggested that Tranilast increased
cell viability and decreased cell death (p<0.05).
