Toll like receptor 9 mRNA expression in calves.

ABSTRACT

We report transcriptional expression of TLR9 in mesenteric lymph nodes, tonsil, spleen, blood, lung and intestine of buffalo calves. The abundance of TLR9 in different tissues was determined by normalizing the integrated density values of TLR9 expression against GAPDH housekeeping gene. TLR9 mRNA expression was validated by RT- PCR. All tissues and peripheral blood mononuclear cells (PBMCs) exhibited variable mRNA expression of TLR9. We found that expression in all tissues and cells differed (p<0.001). Least integrated density values occurred in PBMCs, whereas highest in tonsil. Quantitative PCR yielded highest TLR9 expression in lymph nodes of 25.4 fold higher, spleen 24.2, tonsils 24.82, lungs 17 and intestine 14.3 determined by ∆∆Ct. However, relative levels of TLR9 mRNA levels were not different. We found higher expression of TLR9 in lymphoid tissues compared to PBMCs, lungs and intestine.
