PCR and
probe based nucleic acid detection are used increasingly
in diagnostics. PSI-BLAST (screening), Clustal
W
(sequence homology), phylogenic analysis and structure
prediction were
used to
show that Taq polymerase has
sequence
similarity with
many of the other bacterial polymerase sharing structural
domains XPG,
DNA polyA and Taqexonuc
superfamilies. The findings
suggested that Taq polymerase
might be replaced and diagnosis
carried out
at isothermal conditions.
