N, Kirvalidze L, Mathematical description of the
dose-dependent pharmacokinetics of Phenobarbital, On J Vet Res 11 (1):75 -88, 2007 Pharmacokinetic
plasma phenobarbital concentration curves in the rabbit after different oral
doses were derived mathematically. Humped ascending branches appeared in the
curves at high doses. Absorption and elimination kinetics were determined using
mathematical successive substitution with correlation and regression analysis
versus rate constants of absorption and elimination, as well as a kinetic order
of elimination. Increased dosing significantly affected the pharmacokinetics of
phenobarbital. The findings suggest a step-wise absorption of phenobarbital
into blood.
Key words: phenobarbital, dose
dependent pharmacokinetics; step-wise absorption, successive joining up of
reservoirs, successive substitution method
