Variability in the intron-2 of leptin gene in Reverine
buffaloes, Onl J Vet Res., 10 (2) : 146-155, 2006. A study on leptin gene was
conducted to detect the variability at nucleotide level in Reverine
buffaloes. The PCR-RFLP of 423 bp amplicon of leptin
gene with Hph I, Sau 3A I and Alu
I revealed monomorphism at these loci in Mehsana, Surti and Nili-Ravi breeds of
buffaloes. However on sequencing the amplicon in these breeds variability at
nucleotide positions 43, 47, 51, 70, 71, 76, 89, 94, 99, 150, 194, 213, 267,
315, 335,411 and 415 was identified. The comparison between buffalo sequences
and other species revealed changes of nucleotides at several positions. The
sequences were submitted in the NCBI gene bank with accession numbers:
DQ497641, DQ831142 and DQ831143. On phylogenetic analysis of nucleic acid
sequences all the buffalo leptin sequences clustered into a monophyletic group.
The bovids clustered clearly from Capra hircus
and Ovis aries
cluster.
