ELISA to differentiate avian influenza virus from virus in vaccinated poultry.

ABSTRACT

We tested an ELISA to differentiate avian influenza virus from that in infected poultry. Viruses were isolated from 10-day-old embryonated chicken eggs sub typed by hemagglutination and neuraminidase-inhibition tests with polyclonal chicken antisera. Viruses were passaged in eggs for allantoic fluids harvested as stock virus. Subtype H9N2 was used for inactivated vaccine. Then, 2 groups of 100 chicks each were vaccinated subcutaneously in neck at day 21 with vaccine or infected intra-nasally with 0.2ml allantoic fluid. One hundred controls were not vaccinated or inoculated virus. Blood for sera was taken at day 42 for commercial ELISA kit assay in u-bottomed 96-well microtiter plates. Hemagglutination inhibition was done in U-bottomed 96-well microtiter plates with 5% chicken erythrocytes. By hemagglutinin test reference sensitivity and specificity of ELISA was 100%. Three weeks after infection or vaccination, sera tested positive but we found no significant differences in viruses between infected and vaccinated poultry.
