Effect of intravenous salinomycin on ECG and serum biochemistry in female sheep

Salinomycin ionophore is used as growth promoter in ruminants and coccidiostat in chickens buta high doses can induce tachycardia, muscle tremor, restlessness, loss of appetite, incoordination, muscular weakness and continual panting. We induced acute toxicity with single 0.5mg/kg intravenous salinomycin in six 33 + 3kg mixed breed sheep. Six controls were injected 10 ml saline. Before and after administration, eletrocardiograms were monitored and blood taken for serum liver enzymes and protein. Within 48h, salinomycin was associated with inappetence, dehydration, hyperventilation, tachycardia, fever, paralysis, nasal discharge, bruxism and death in 1 sheep. Compared with controls, serum aspartase increased 32-55% 24h after injection, alanine transaminase 46-59% by 7-14 days, lactate dehydrogenase 31-76 within 24h and creatinine kinase 44-75% 7-14 days (P < 0.05). By auscultation, mean heart rate increased 43% and by electrocardiogram 42% (P < 0.05). In controls we recorded 3 sinus tachycardia and 2 arrhythmias whereas in those given salinomycin found 7 tachicardias 4 supraventricular tachycardia, 2 supra ventricular premature contraction and 3 wavw inversions. Our findings suggest single IV high dose of salinomycin may induce severe liver and cardiac toxicity.
