Mutations
in mitochondria (mtDNA) can affect the incidence and
severity of disease and abnormalities in vertebrates. Characterization and sequencing of the ND5 gene in Khorasan native
poultry is reported. After extraction, two DNA fragments were amplified by a
specific primer, cloned to vector pTZ57R/T and transformed to an E. coli -
DH5α strain. Recombinant plasmids were extracted from positive colonies
and sequenced. Results showed maximum similarity with Gallus Gallus mitochondrial genome and a
protein BLAST query showed that the sequence had a similarity of 93% in 95%
coverage to NADH dehydrogenase subunit 5. The protein BLAST query suggested the
conserved domain as NADH5 C and Oxidored q1, in a family
which is part of a complex I. This catalyzes the transfer of two electrons from
NADH to ubiquinone in a reaction associated with proton translocation across
the membrane. The findings may be useful in identifying phenotypes in poultry.
