Early diagnosis of brucellosis in goats.

ABSTRACT

Baladi does were inoculated Brucella melitensis biovar 3 at 40-45 days gestation and blood taken every 7 days. Inoculated does showed severe respiratory disorders and abortion by 3 – 5 weeks. Br. melitensis was isolated from the spleen, and lymph nodes from all inoculated does. For antibody we used ELISA, plate antigen (BAPAT), rose bengal (RBPT), rivanol (RVT), complement (CFT) and agglutination (TAT) tests.  ELISA, BAPAT, RBPT and RVT tests detected antibody by 4 to 7 days but CFT and TAT by 14th day, peaking by 1st week for BAPAT and RBPT, 3-4th for TAT and RVT and 5th week for CFT and ELISA. Antibodies remained high 18 weeks by BAPAT and RBPT, 16 for RVT, 13 by ELISA and 11 weeks for TAT and CFT. Compared with controls, in inoculated animals, we found a ~7-10 fold boost in lymphocyte transformations. By microscopy we observed diffuse suppurative and necrotizing placentitis, and ulcerative endometritis with large colonies of intra and extra cellular Brucella. Findings suggest RBPT should be combined with ELISA while CFT and/or PCR as confirmatory tests.
