Colony-PCR for direct extraction of DNA from Corynebacterium pseudotuberculosis.

ABSTRACT

 

Taha Yassin, Ghani Al-Badrawi, Colony PCR for direct extraction of DNA from Corynebacterium pseudotuberculosis, Onl J Vet Res., 23 (12):1184-1187, 2019. Authors describe a method to extract DNA directly from Corynebacterium pseudotuberculosis for PCR. C. pseudotuberculosis was cultured anaerobically on solid media incubated at 37C for 48-72 hours. C. pseudotuberculosis colony was added to PCR master mix mixture instead of DNA template extract. We then amplified the PCR product on agarose gel along with a DNA marker producing clear wide compact electrophoretic DNA bands. We find this PCR assay could identify Corynebacterium pseudotuberculosis directly without the need to extract DNA from bacterial isolates.
